I have a 6×7 ft room with 8ft cieling I have a fan and a large dehumidifier the room is well insulated and sealed It has 1 3×6" vent in high on the wall , the 3 plants in there are doing well still in veg stage , my question is how crucial is it to get fresh air into the room is my 1 small vent enough?

You might want to stagger those during flower if you can...like a few weeks in between each plant. Will help with crowdedness, humidity, etc.

Sealed room needs co2 enrichment. A room with a vent isn’t sealed.
The one vent has no fan? Are you attempting to pull air out or pushing air through? You will need plenty of fresh air, but might be difficult for the dehumidifier to keep up with the fresh air pulled in.
